Contains HieffTM Taq DNA Polymerase (Cat#10101ES), dNTPs, and other PCR-required components. The Master Mix is stable for 3 months at 4℃ with our customized stabilizers. The pre-mix solution is optimized for conventional PCR and ready to use by adding DNA template and primers. The PCR products can be loaded directly for electrophoresis with pre-loaded bromophenol blue dye. The amplified products contain 3 '-dA protrusion and can be easily cloned into T vector. 2× Hieff™ PCR Master Mix (No Dye) - 10103ES
Conventional PCR premixed solution which is ready to use, including HieffTM Taq DNA Polymerase (Cat#10101), dNTP mix, MgCl2 and optimized buffer. During the reaction, only the primer and template can be added for amplification, which greatly simplifies the operation steps of experiment. This product contains excellent stabilizers and can be stored for 3 months at 4℃. The PCR product have 3 '-dA protrusion and can be easily cloned into T vector.

Hieff™ Taq DNA Polymerase - 10101ES
A thermostable recombinant wild type DNA polymerase expressed by Thermus aquaticus with a molecular weight of 94 KDa. It has 5'→3' polymerase activity and 5'→3' exonuclease activity but without 3'→5' exonuclease activity. The amplified product has 3'- dA and can be directly used for TA cloning.

PCR Enhancer - 10117ES
PCR Enhancer is a mixed additive composed of multiple components. In PCR reaction, PCR Enhancer can effectively reduce the melting temperature of high GC templates and templates with complex secondary structure, and greatly reduce the influence of DNA secondary structure on PCR, which increases the sensitivity and specificity of PCR reaction, and is compatible with almost all DNA polymerase amplification systems. When the optimized PCR program cannot effectively amplify the target fragment, adding PCR Enhancer can often get excellent results. This product may reduce PCR fidelity. Therefore, use it with caution when performing high-fidelity PCR.

Ready-to-use 2× pre-mixed solution containing Hieff Canace™ AdvanceFast High-Fidelity DNA Polymerase, dNTPs, and an optimized buffer system, which contains pre-added electrophoresis indicators. The pre-mix contains pre-added electrophoresis indicator, PCR products can be directly electrophoresed, the amplification products are flat ends. 2× Hieff Canace™ AdvanceFast PCR Master Mix (With Dye) has the advantages of quick and easy, high sensitivity, high specificity, good stability, etc., the reaction system can be added with only the primers and templates. In addition, the product also contains a specific protective agent, so that the premix can still maintain stable activity after repeated freezing and thawing.

The assay was done by examining the mutation frequency of PCR products of 6 GC-rich (about 80%) segments amplified from the genomes of different species. The PCR products amplified by Hieff Canace™ AdvanceFast High-Fidelity DNA Polymerase were cloned into vectors, and 100 selected single clones for each sequence were subjected to Sanger sequencing.

Hieff Canace™ Plus High-Fidelity DNA Polymerase - 10153ES
Hieff Canace™ Plus High-Fidelity DNA Polymerase is based on the Pyroccus Furiosis DNA Polymerase, genetically engineered. The enzyme has a 5'→3' DNA polymerase activity and a 3'→5' exonuclease activity, and its fidelity is 83 times that of Taq DNA polymerase and 9 times that of ordinary Pfu DNA polymerase. Two monoclonal antibodies at room temperature that inhibit polymerase activity and 3'→5' exonuclease activity were added to the enzyme solution, which can easily perform highly specific Hot Start PCR, greatly improving the detection rate of amplification and the specificity of the product. The addition of an extension factor to the enzyme solution gives the enzyme the ability to amplify long fragments, and the length of the amplification of the fragment of interest can be up to 13 kb. This product is equipped with an optima buffer that makes the enzyme suitable for amplification of complex templates. It generates blunt ends in the amplification products.

Genotyping PCR
2×Hieff® HotStart PCR Genotyping Master Mix(With Dye) - 10108ES
Ready-to-use PCR premixed solution containing HieffTM HotStart Taq DNA Polymerase, dNTPs, and an optimized buffering system. Simply add primers and template for amplification, which greatly simplifies the experimental procedure and allows for high-throughput operations, enhancing the reproducibility of results. HieffTM HotStart Taq DNA Polymerase is a ligand-modified thermostable Taq DNA Polymerase, with a ligand that modulates the activity of the DNA polymerase in response to temperature changes. The enzyme activity is completely sealed at room temperature and is only released after heating at 95℃. The activation time for HieffTM HotStart DNA Polymerase requires only 2-3 min and is compatible with existing PCR protocols. This product prevents non-specific amplification during sample preparation and reaction heating stages, effectively enabling genotyping experiments.

Mouse Tissue/ Cell Lysis Reagent - 19697ES
The kit is equipped with a powerful lysis buffer, which can quickly lyse samples (e.g. cells, mouse tail, mouse ear, mouse toe, muscle, etc.) to release genomic DNA, and the lysate can be directly added to the PCR reaction system without purification, which is convenient for operation. In addition, this kit requires low sample input, 5 mg of mouse tissue or 1-5 mm of mouse tail can be used for experiments.
Instructions for Mouse Genomic DNA Release
- Clip 5-10 mg of animal tissue or 1-5 mm mouse tail in a 1.5 mL centrifuge tube*.
- Add 90 μL of Buffer ML to the above centrifuge tube, vortex gently to completely infiltrate the sample with the lysate, and centrifuge briefly.
- Incubate at 95°C for 15 min in a thermostatic incubator**.
- Add 10 μL of Buffer MT, flick to mix, and terminate lysis.
- Optional step: centrifugation at 12000 rpm for 2 min.
- Transfer the supernatant to a new centrifuge tube and store at 4°C or -20°C or take the supernatant directly for subsequent PCR amplification.
Long PCR
2×Hieff® Ultra-Rapid II HotStart PCR Master Mix - 10167ES
The mix contains heat-stabilized Taq DNA Polymerase modified with antibodies, adds strong elongation factor and optimized buffer system, and has super high amplification efficiency. It is very suitable for PCR amplification of most colonies such as E. coli, Agrobacterium, and yeast. The extension speed of amplification of complex templates within 3 kb can reach 1 sec/kb, 3-6 kb to 3 sec/kb, 6-10 kb to 5 sec/kb and 10 kb to 10 sec/kb; the amplification speed of simple templates such as plasmid within 6 kb can reach 1 sec/kb, which can greatly save PCR reaction time. Meanwhile, Mix contains dNTPs, Mg2+, and it can be amplified only with adding primers and template. In addition, Mix contains red tracer dye, which can be used by electrophoresis directly after the end of the reaction. The protective agent added to the system enables the product to maintain a stable activity after repeated freezing and thawing. The 3′ end of the PCR product bands A and can be easily cloned into the T vector.

Blood Advanced Direct PCR Kit - 10188ES
The Blood Advanced Direct PCR Kit is a kit that enables direct PCR amplification of whole blood samples without DNA purification or sample pretreatment. This kit is compatible with fresh blood containing conventional anticoagulants such as EDTA, heparin, citrate, frozen blood and commercial Whatman903TM and FTATM dried blood stains. The kit contains a combination of genetically engineered DNA Polymerase with high fidelity and tolerance to PCR inhibitors. It can efficiently amplify 8 kb genomic fragments under rapid elongation conditions, for the fragments less than 2 kb, the extension time of 3-5 sec/kb can be set to complete the amplification, thus significantly reducing the identification and detection time of blood samples.
HieffTM Advanced High-Fidelity DNA Polymerase Mix has a flat 3 'end of the amplified product, which is suitable for one-step rapid cloning and TOPO cloning (YEASEN Cat#10909/10910/10911/10922).
The Positive control primer mix (10 μmol/L each) provided in the kit is capable of amplifying a fragment 237 bp in length from the upstream conserved sequence of the sox21 gene in mammals and most vertebrates, which could be used as a positive control.

Plant Tissue Direct PCR Kit (With Dye) - 10187ES
Plant tissue direct PCR kit is a kit that can directly amplify different types of plant leaves by PCR, with wide adaptability and strong stability. The kit uses a unique lysis buffer system, which can quickly lyse a variety of plant samples and release genomic DNA. The released genomic DNA can be used directly as a template without removing protein, RNA, or secondary metabolites in PCR reaction. In addition, the kit requires a small amount of sample, as low as 1 mm plant leaves can be used for experiments.

Recommendations:
- When doing leaf experiments, it is recommended to use freshly collected leaf tissue. If it is a long-term frozen tissue, it needs to be stored at -80°C. Repeated freezing and thawing should be avoided as much as possible to avoid template degradation and affect PCR efficiency. The leaf tissue is suitable for young leaves. If it is a mature leaf, avoid using the tissue of the main vein of the leaf
- It is recommended to amplify the fragment within 1 kb in length for the best amplification efficiency
- When sampling, use a hole punch or knife to take a sample of suitable size. When the samples are different, the hole punch or knife needs to be cleaned every time before processing the sample
- For leaf tissue, it is recommended to take 1-10 mm leaves, too small leaf length will lead to low PCR amplification yield, too much will inhibit the PCR reaction, use the method of thermal cracking, mashing with a pipette tip, and crushing with a grinder to treat plant leaves. After treatment, it needs to be shaken and centrifuged. Be sure to take the supernatant for testing. Precipitation will seriously inhibit the PCR reaction.
